Over expression of the cytosolic form of the metabolic enzyme PhosphoEnolPyruvate Carboxykinase (PEPCK-Cmus) in the skeletal muscle has led to the creation of a line of super mice. This enzyme has been discovered to be involved in two main pathways, Gluconeogenesis and Glyceroneogenesis. These mice have the capability of running five to six kilometers continuously at a speed of 20 meters per minute on a treadmill. Even though, they consume, on average, 60% more food, they weigh 50% less than their wildtype control. They exhibit an increase in life expectancy and are capable of breeding at older ages as well as they are capable to perform high endurance behavioral tasks longer than their control wildtypes. We have carried out many experiments on theses genetically enhanced mice to obtain more data, such as the level of lactate and glucose changes during exercise and the hormonal changes before and after exercise. These mice have raised more questions than answers about PhosphoEnolPyruvate Carboxykinase. However, this research hopefully would be able to help us get a better insight to the exact roll of the PEPCK gene and enzyme.
